影片以悲壯的筆觸，描述了30年代時期，不丹國卡爾城金將軍與日軍勾結，在城里生產毒氣，中國派遣特務“天字第一號”和現代鏢局負責接出城主尤達。女鏢師阿瓊在愛慕尤達的金將軍女兒芊芊幫助下，意欲和尤達一起逃走，但“天字第一號”力主要破壞毒氣計劃，結果被抓。金將軍為救愛女，反過來幫助他們，與日軍開始一場大戰。

　　As bacteriologist Lena (38) drives past a burning car, she hears a scream of a dying woman. 　　香港警察朱福龍（甄子丹 飾）因一次工作失誤慘被降級，萬般無奈之際又遭未婚妻可兒（周勵淇 飾）悔婚，事業愛情的雙重打擊令他一蹶不振，暴飲暴食六個月后體重猛增到220磅！上司黃警官不忍其繼續墮落，委派朱福龍前往日本押送一名犯人，并承諾只要完成任務就可以為他恢復職位。可惜來到日本，倒霉的事件便接連在他身上發生，不僅財物盡失，連犯人也在押送途中逃脫了。幸好得到前香港警察瀟灑哥（王晶 飾）的幫助才得以暫時解困，為了完成任務找回犯人，朱福龍與瀟灑哥共同行動，不料在查找真相的過程中發現了日本黑幫的驚天陰謀......
